Therapie
Antineoplastika > Andere Antineoplastika > PARP-Inhibitoren
Indikation
Lokal fortgeschrittenes oder metastasiertes HER2-negatives Mammakarzinom mit gBRCA-Mutation nach Vorbehandlung mit einem Anthracyclin und/oder einem Taxan (bei Hormonrezeptor-positivem Brustkrebs: nach Progression unter endokriner Therapie oder wenn eine endokrine Therapie ungeeignet ist).
Befristet zugelassene Indikation: asymptomatisches oder leicht symptomatisches metastasiertes, kastrationsresistentes Prostatakarzinom mit Mutationen in homologen Rekombinationsreparaturgenen in Kombination mit Enzalutamid, wenn eine Chemotherapie nicht indiziert ist.
Befristet zugelassene Indikation: asymptomatisches oder leicht symptomatisches metastasiertes, kastrationsresistentes Prostatakarzinom mit Mutationen in homologen Rekombinationsreparaturgenen in Kombination mit Enzalutamid, wenn eine Chemotherapie nicht indiziert ist.
Dosierung
Unabhängig der Mahlzeiten.
Mammakarzinom: >18 J.: 1×tgl. 1 mg.
Prostatakarzinom (befristet zugelassene Indikation): >18 J.: 1×tgl. 0,5 mg in Kombination mit Enzalutamid 1×tgl. 160 mg (und einem GnRH-Analogon bei Patienten ohne bilaterale Orchiektomie).
Mammakarzinom: >18 J.: 1×tgl. 1 mg.
Prostatakarzinom (befristet zugelassene Indikation): >18 J.: 1×tgl. 0,5 mg in Kombination mit Enzalutamid 1×tgl. 160 mg (und einem GnRH-Analogon bei Patienten ohne bilaterale Orchiektomie).
Kontraindikation
Schwangerschaft (bis min. 7 Mon. nach Therapie), Fortpflanzung beim Mann und Geschlechtsverkehr ohne Kondom bei Männern mit schwangeren Partnerinnen (bis min. 4 Mon. nach Therapie), Stillzeit (bis min. 1 Mon. nach Therapie).
